This assay has high sensitivity and excellent specificity for detection of human alpha-SMA. No significant cross-reactivity or interference between human alpha-SMA and analogues was observed.

Samples: Serum, tissue homogenates, urine. Assay Type: Sandwich. Detection Range: 3.12 ng/ml -200 ng/ml. Sensitivity: 0.78 ng/ml.

Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.

Principle of the Assay This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for alpha-SMA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any alpha-SMA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for alpha-SMA is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of alpha-SMA bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene belongs to the actin family of proteins, which are highly conserved proteins that play a role in cell motility, structure and integrity. Alpha, beta and gamma actin isoforms have been identified, with alpha actins being a major constituent of the contractile apparatus, while beta and gamma actins are involved in the regulation of cell motility. This actin is an alpha actin that is found in skeletal muscle. Defects in this gene cause aortic aneurysm familial thoracic type 6.
