For Sample For the quantitative determination of mouse tumor necrosis factor ? (TNF-?) concentrations in serum, plasma, cell culture supernates, tissue homogenates. PRINCIPLE OF THE ASSAY This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TNF-? has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TNF-? present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNF-? is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNF-? bound in the initial step. The color development is stopped and the intensity of the color is measured.

Members of this family are classified based on primary sequence, function, and structure. This protein is synthesized as a type-II transmembrane protein and is reported to be cleaved into products that exert distinct biological functions. It plays an important role in the innate immune response as well as regulating homeostasis but is also implicated in diseases of chronic inflammation. In mouse deficiency of this gene is associated with defects in response to bacterial infection, with defects in forming organized follicular dendritic cell networks and germinal centers, and with a lack of primary B cell follicles.
