For samples: For the quantitative determination of human protein Z concentrations in serum, plasma, tissue homogenates. PRINCIPLE OF THE ASSAY This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for protein Z has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any protein Z present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for protein Z is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of protein Z bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a liver vitamin K-dependent glycoprotein that is synthesized in the liver and secreted into the plasma. The encoded protein plays a role in regulating blood coagulation by complexing with protein Z-dependent protease inhibitor to directly inhibit activated factor X at the phospholipid surface. Deficiencies in this protein are associated with an increased risk of ischemic arterial diseases and fetal loss. Mutations in this gene are the cause of protein Z deficiency.
