Samples: Serum, plasma and urine. Assay Type: Competitive. Detection Range: 0.1 ng/ml-40 ng/ml. Sensitivity: The minimum detectable dose of human NMP-22 is typically less than 0.04ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest human NMP-22 concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.

Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<6%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<11%. Three samples of known concentration were tested in twenty assays to assess.

Principle of the assay: This assay employs the competitive inhibition enzyme immunoassay technique. Antibody specific for NMP-22 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells with a Horseradish Peroxidase (HRP) conjugated NMP-22. A competitive inhibition reaction is launched between NMP-22 (Standards or samples) and HRP-conjugated NMP-22 with the pre-coated antibody specific for NMP-22. The more amount of NMP-22 in samples, the less antibody bound by HRP-conjugated NMP-22. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in opposite to the amount of NMP-22 in the sample. The color development is stopped and the intensity of the color is measured.
