Introduction: Pregnancy-associated plasma protein A, pappalysin 1, also known as PAPP-A, is a protein used in screening tests for Down syndrome. This protein is a secreted metalloproteinase which cleaves insulin-like growth factor binding proteins (IGFBPs). It is thought to be involved in local proliferative processes such as wound healing and bone remodeling. Low plasma level of this protein has been suggested as a biochemical marker for pregnancies with aneuploid fetuses (fetuses with an abnormal number of chromosomes). For example, low PAPPA may be seen in prenatal screening for Down syndrome. Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. An antibody specific for PAPP-A antibody has been pre-coated onto a microplate. A competitive inhibition reaction is launched between HRP labeled PAPP-A and unlabeled PAPP-A (Standards or samples) with the antibody specific for PAPP-A. The substrate solutions are added to the wells, 3 respectively. And the color develops in opposite to the amount of PAPP-A bound in the initial step. The color development is stopped and the intensity of the color is measured.
