Princple of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for NCAM-L1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NCAM-L1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NCAM-L1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NCAM-L1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is an axonal glycoprotein belonging to the immunoglobulin supergene family. The ectodomain, consisting of several immunoglobulin-like domains and fibronectin-like repeats (type III), is linked via a single transmembrane sequence to a conserved cytoplasmic domain. This cell adhesion molecule plays an important role in nervous system development, including neuronal migration and differentiation. Mutations in the gene cause X-linked neurological syndromes known as CRASH (corpus callosum hypoplasia, retardation, aphasia, spastic paraplegia and hydrocephalus).
