Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. Antibody specific for HAS has been pre-coated onto a microplate. Standards and samples are pipetted into the wells with a Horseradish Peroxidase (HRP) conjugated HAS. A competitive inhibition reaction is launched between HAS (Standards or samples) and HRP-conjugated HAS with the pre-coated antibody specific for HAS. The more amount of HAS in samples, the less antibody bound by HRP-conjugated HAS. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in opposite to the amount of HAS in the sample. The color development is stopped and the intensity of the color is measured.

Albumin is a soluble, monomeric protein which comprises about one-half of the blood serum protein. Albumin functions primarily as a carrier protein for steroids, fatty acids, and thyroid hormones and plays a role in stabilizing extracellular fluid volume. Albumin is a globular unglycosylated serum protein of molecular weight 65,000. Albumin is synthesized in the liver as preproalbumin which has an N-terminal peptide that is removed before the nascent protein is released from the rough endoplasmic reticulum.
