Introduction: Mucins are a family of high molecular weight, heavily glycosylated proteins (glycoconjugates) produced by many epithelial tissues in vertebrates. They bind to pathogens as part of the immune system. Overexpression of the mucin proteins, especially MUC1 is associated with many types of cancer. Although some mucins are membrane-bound due to the presence of a hydrophobic membrane-spanning domain that favors retention in the plasma membrane, most mucins are secreted onto mucosal surfaces or secreted to become a component of saliva. At least 19 human mucin genes have been distinguished by cDNA cloning-MUC1, MUC2, MUC3A, MUC3B, MUC4, MUC5AC, MUC5B, MUC6, MUC7, MUC8, MUC12, MUC13, MUC15, MUC16, MUC17, MUC19, and MUC20. The major secreted airway mucins are MUC5AC and MUC5B, while MUC2 is secreted mostly in the intestine but also in the airway. The human mucin gene MUC5AC is mapped clustered with MUC2, MUC5B and MUC6 on chromosome 11p15.5. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to MUC5AC. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for MUC5AC and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain MUC5AC, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of MUC5AC in the samples is then determined by comparing the O.D. of the samples to the standard curve.
