. Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for FVIII has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FVIII present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FVIII is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FVIII bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes coagulation factor VIII, which participates in the intrinsic pathway of blood coagulation; factor VIII is a cofactor for factor IXa which, in the presence of Ca+2 and phospholipids, converts factor X to the activated form Xa. This gene produces two alternatively spliced transcripts. Transcript variant 1 encodes a large glycoprotein, isoform a, which circulates in plasma and associates with von Willebrand factor in a noncovalent complex. This protein undergoes multiple cleavage events. Transcript variant 2 encodes a putative small protein, isoform b, which consists primarily of the phospholipid binding domain of factor VIIIc.
