Introduction: Nitric oxide synthases (NOSs) are present among eukaryoticenzymes as dimeric, calmodulin-dependent orcalmodulin-containing cytochrome p450-like hemoprotein thatcombine reductase and oxygenase catalytic domains in onedimer, bear both flavin adenine dinucleotide (FAD) and flavinmononucleotide (FMN), and carry out a 5`-electron oxidation ofnon-aromatic amino acid L-arginine with the aid oftetrahydrobiopterin. NOS is an enzyme in the body thatcontributes to transmission from one neuron to another, to theimmune system and to dilating blood vessels. It does so bysynthesis of nitric oxide from the terminal nitrogen atom ofL-arginine in the presence of NADPH and dioxygen. NOS is theonly known enzyme that binds FAD, FMN, heme,tetrahydrobiopterin (BH4) and calmodulin.There are three known isoforms, two are constitutive (cNOS) andthe third is inducible (iNOS). As opposed to the criticalcalcium-dependent regulation of constitutive NOS enzymes(nNOS and eNOS), iNOS has been described ascalcium-insensitive, likely due to its tight non-covalent interactionwith calmodulin and Ca2+. While evidence for 'baseline' iNOS expression has been elusive, IRF-1 and NF-kappaB-dependentactivation of the inducible NOS promoter supports aninflammation mediated stimulation of this transcript. From afunctional perspective, it is important to recognize that inductionof the high-output iNOS usually occurs in an oxidativeenvironment, and thus high levels of NO have the opportunity toreact with superoxide leading to peroxynitrite formation and celltoxicity. These properties may define the roles of iNOS in hostimmunity, enabling its participation in anti-microbial andanti-tumor activities as part of the oxidative burst ofmacrophages. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated withan antibody specific to iNOS. Standards or samples are thenadded to the appropriate microtiter plate wells with abiotin-conjugated antibody preparation specific for iNOS andAvidin conjugated to Horseradish Peroxidase (HRP) is added toeach microplate well and incubated. Then a TMB (3,3',5,5'tetramethyl-benzidine) substrate solution is added to each well.Only those wells that contain iNOS, biotin-conjugated antibodyand enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of asulphuric acid solution and the color change is measuredspectrophotometrically at a wavelength of 450 nm +/- 2 nm. Theconcentration of iNOS in the samples is then determined bycomparing the O.D. of the samples to the standard curve.
