Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique.Antibody specific for AQP-1 has been pre-coated onto a microplate. Standardsand samples are pipetted into the wells and any AQP-1 present is bound by theimmobilized antibody. After removing any unbound substances, abiotin-conjugated antibody specific for AQP-1 is added to the wells. Afterwashing, avidin conjugated Horseradish Peroxidase (HRP) is added to thewells. Following a wash to remove any unbound avidin-enzyme reagent, asubstrate solution is added to the wells and color develops in proportion to theamount of AQP-1 bound in the initial step. The color development is stoppedand the intensity of the color is measured.

Aquaporins are a family of small integral membrane proteins related to the major intrinsic protein (MIP or AQP0). This gene encodes an aquaporin which functions as a molecular water channel protein. It is a homotetramer with 6 bilayer spanning domains and N-glycosylation sites. The protein physically resembles channel proteins and is abundant in erythrocytes and renal tubes. The gene encoding this aquaporin is a possible candidate for disorders involving imbalance in ocular fluid movement.
