Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for C4b has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any C4b present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for C4b is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of C4b bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein is expressed as a single chain precursor which is proteolytically cleaved into a trimer of alpha, beta, and gamma chains prior to secretion. The trimer provides a surface for interaction between the antigen-antibody complex and other complement components. The alpha chain may be cleaved to release C4 anaphylatoxin, a mediator of local inflammation. Deficiency of this protein is associated with systemic lupus erythematosus. This gene localizes to the major histocompatibility complex (MHC) class III region on chromosome 6. Varying haplotypes of this gene cluster exist, such that individuals may have 1, 2, or 3 copies of this gene.
