Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for LILRB4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any LILRB4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LILRB4 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LILRB4 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene is a member of the leukocyte immunoglobulin-like receptor (LIR) family, which is found in a gene cluster at chromosomal region 19q13.4. The encoded protein belongs to the subfamily B class of LIR receptors which contain two or four extracellular immunoglobulin domains, a transmembrane domain, and two to four cytoplasmic immunoreceptor tyrosine-based inhibitory motifs (ITIMs). The receptor is expressed on immune cells where it binds to MHC class I molecules on antigen-presenting cells and transduces a negative signal that inhibits stimulation of an immune response. The receptor can also function in antigen capture and presentation. It is thought to control inflammatory responses and cytotoxicity to help focus the immune response and limit autoreactivity.
