Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with biotin-BSA and Avidin. Standards and samples are pipetted into the wells with a biotin-conjugated antibody specific for GHBP. After washing, GHBP antibody conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GHBP in sample. The color development is stopped and the intensity of the color is measured.

Binding of growth hormone to the receptor leads to receptor dimerization and the activation of an intra- and intercellular signal transduction pathway leading to growth. Mutations in this gene have been associated with Laron syndrome, also known as the growth hormone insensitivity syndrome (GHIS), a disorder characterized by short stature. In humans and rabbits, but not rodents, growth hormone binding protein (GHBP) is generated by proteolytic cleavage of the extracellular ligand-binding domain from the mature growth hormone receptor protein.
