This assay has high sensitivity and excellent specificity for detection of rat TAFI. No significant cross-reactivity or interference between rat TAFI and analogues was observed.

Samples: For the quantitative determination of rat thrombin activatable fibrinolysis inhibitor (TAFI) concentrations in serum, plasma, tissue homogenates. Detection Range: 3.125 ng/ml -200 ng/ml.

Sensitivity: The minimum detectable dose of rat TAFI is typically less than 1.57 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest rat TAFI concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations. Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.

Principle of the assay: </b<This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with TAFI. Standards or samples are added to the appropriate microtiter plate wells with Horseradish Peroxidase (HRP) conjugated antibody preparation specific for TAFI. The competitive inhibition reaction is launched between with pre-coated TAFI and TAFI in samples. A substrate solution is added to the wells and the color develops in opposite to the amount of TAFI in the sample. The color development is stopped and the intensity of the color is measured.
