Introduction: The zona pellucida is a glycoprotein membrane surrounding theplasma membrane of an oocyte. It is a vital constitutive part of thelatter, external but not extraneous to it. The zona pellucida firstappears in multilaminar primary oocytes. This structure bindsspermatozoa, and is required to initiate the acrosome reaction. Autoantibodies to zona-pellucida seem to be important autoantibodiesimplicated in reproduction, with substantial role in both endocrine andreproductive functions of the human ovary. Autoantibodies in serumof female patients directed against antigens of the zona pellucida,antizona pellucida antibodies or zona pellucida antibodies, are crucialfor female infertility. Pregnancy rates of women with antizonapelludica antibodies are drastically reduced even after an assistedreproduction therapy. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated withspecificity antigen. Samples are then added to the appropriatemicrotiter plate wells and incubated. Then add HorseradishPeroxidase (HRP)-conjugated anti-human IgG and incubate again.Substrate solutions are added to each well. The enzyme-substratereaction is terminated by the addition of a sulphuric acid solution andthe color change is measured spectrophotometrically at a wavelengthof 450 nm +/- 2 nm. Calculate the valence of anti-zona pellucidaantibody in the samples.
