This assay has high sensitivity and excellent specificity for detection of Human S100A14. No significant cross-reactivity or interference between Human S100A14 and analogues was observed.

Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV% is less than 8%. Three samples of known concentration were tested twenty times on one plate to assess. Introduction: S100A14 is a member of a subfamily of proteins related by Ca(2+)-binding motifs to the EF-hand Ca(2+)-binding protein superfamily. Expressed at highest levels in colon and at moderate levels in thymus, kidney, liver, small intestine, and lung. S100A14 is also overexpressed in ovary, breast and uterus tumours . Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to S100A14. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for S100A14 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain S100A14, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of S100A14 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a member of the S100 protein family which contains an EF-hand motif and binds calcium. The gene is located in a cluster of S100 genes on chromosome 1. Levels of the encoded protein have been found to be lower in cancerous tissue and associated with metastasis suggesting a tumor suppressor function (PMID: 19956863, 19351828). Function: Modulates P53/TP53 protein levels, and thereby plays a role in the regulation of cell survival and apoptosis. Depending on the context, it can promote cell proliferation or apoptosis. Plays a role in the regulation of cell migration by modulating the levels of MMP2, a matrix protease that is under transcriptional control of P53/TP53.
