Introduction: Neurotrophins are a family of proteins that induce the survival, development and function of neurons. They belong to a class of growth factors, secreted proteins, which are capable of signaling particular cells to survive, differentiate, or grow. Growth factors such as neurotrophins that promote the survival of neurons are known as neurotrophic factors. Neurotrophic factors are secreted by target tissue and act by preventing the associated neuron from initiating programmed cell death - thus allowing the neurons to survive. Neurotrophins also induce differentiation of progenitor cells, to form neurons. Neurotrophin-4 (NT-4) like BDNF, is a neurotrophic factor that signals predominantly through the TrkB receptor tyrosine kinase. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to NT-4. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for NT-4 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain NT-4, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of NT-4 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
