Direct Detection: To detect Human sTRAIL/Apo2L by direct ELISA (using 100 mul/well antibody solution) a concentration of 0.25 - 1.0 mug/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with compatible secondary reagents, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Human sTRAIL/Apo2L. Sandwich ELISA: To detect Human sTRAIL/Apo2L by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.25 - 1.0 mug/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with Polyclonal Anti-Human sTRAIL/Apo2L as a capture antibody, allows the detection of at least 2000-4000 pg/ml of recombinant Human sTRAIL/Apo2L. Western Blot: To detect Human sTRAIL/Apo2L by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Human sTRAIL/Apo2L is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions. Optimal dilutions should be determined by each laboratory for each application.

The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This protein preferentially induces apoptosis in transformed and tumor cells, but does not appear to kill normal cells although it is expressed at a significant level in most normal tissues. This protein binds to several members of TNF receptor superfamily including TNFRSF10A/TRAILR1, TNFRSF10B/TRAILR2, TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, and possibly also to TNFRSF11B/OPG. The activity of this protein may be modulated by binding to the decoy receptors TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4, and TNFRSF11B/OPG that cannot induce apoptosis. The binding of this protein to its receptors has been shown to trigger the activation of MAPK8/JNK, caspase 8, and caspase 3.
