The lyophilized antibody is stable for at least 2 years from date of receipt at 20 degree C. The reconstituted antibody is stable for at least two weeks at 2 - 8 degree C. Frozen aliquots are stable for at least 6 months when stored at 20 degree C.
tested application :
ELISA (EIA), Western Blot (WB), Immunohistochemistry (IHC)
app notes :
Immunohistochemistry: This antibody stained formalin - fixed, paraffin - embedded sections of human breast invasive ductal carcinoma. The recommended concentration is 2.5 ug/ml and a two - hour incubation at room temperature. An HRP - labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6.0 Sodium Citrate buffer is recommended. Optimal concentrations and conditions may vary. ELISA: To detect human MCP - 1/MCAF by sandwich ELI SA (using 100 ul/well antibody s olution) a concentration of 0.5 – 2.0 ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant human MCP 1/MCA F. Western Blot: To detect h MCP - 1/MCAF by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant human MCP - 1/MCAF is 1.5 - 3.0 ng/lane, under either reducing or non - reducing conditions
other info1 :
Antibody Generation: Produced from sera of rabbits pre-immunized with highly pure (>98%) recombinant hMCP 1/MCAF (human Macrophage/Monocyte chemotactic protein-1). Anti-hMCP-1/MCAF specific antibody was purified by affinity chromatography employing immobilized hMCP-1/MCAF matrix. Immunogen: Recombinant Human MCP-1/MCP-1/MCAF.
other info2 :
Buffer: Water. Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of human MCP-1/MCAF (10.00 ng/ml), a concentration of 3.0 - 5.0 ug/ml of this antibody is required. Reconstitution: Reconstitute the antibody in sterile water to a concentration of 0.1 - 1.0 mg/ml .
products description :
The MCP proteins belong to the CC chemokine family, and signal through CCR2 and, with the exception of MCP-1, other CCR receptors. The MCP proteins chemoattract and activate monocytes, activated T cells, basophils, NK cells, and immature dendritic cells. The MCP family cross reacts across species. Recombinant human MCP-1 is an 8.6 kDa protein containing 76 amino acid residues including the four highly conserved cysteine residues present in the CC chemokines.

ncbi summary :
This gene is one of several cytokine genes clustered on the q-arm of chromosome 17. Chemokines are a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of N-terminal cysteine residues of the mature peptide. This chemokine is a member of the CC subfamily which is characterized by two adjacent cysteine residues. This cytokine displays chemotactic activity for monocytes and basophils but not for neutrophils or eosinophils. It has been implicated in the pathogenesis of diseases characterized by monocytic infiltrates, like psoriasis, rheumatoid arthritis and atherosclerosis. It binds to chemokine receptors CCR2 and CCR4. uniprot summary :
Function: Chemotactic factor that attracts monocytes and basophils but not neutrophils or eosinophils. Augments monocyte anti-tumor activity. Has been implicated in the pathogenesis of diseases characterized by monocytic infiltrates, like psoriasis, rheumatoid arthritis or atherosclerosis. May be involved in the recruitment of monocytes into the arterial wall during the disease process of atherosclerosis. Subunit structure: Monomer or homodimer; in equilibrium. Binds to CCR2 and CCR4. Is tethered on endothelial cells by glycosaminoglycan (GAG) side chains of proteoglycans. Ref.20 Ref.22. Subcellular location: Secreted. Induction: Up-regulated upon hypertonic conditions. Ref.26. Post-translational modification: Processing at the N-terminus can regulate receptor and target cell selectivity. Deletion of the N-terminal residue converts it from an activator of basophil to an eosinophil chemoattractant. Polymorphism: Genetic variations in CCL2 determine Mycobacterium tuberculosis susceptibility [. MIM:607948]. Sequence similarities: Belongs to the intercrine beta (chemokine CC) family.
