Sandwich ELISA: To detect human PF-4 by Sandwich ELISA (using 100 ul/well) a concentration of 0.5–2 ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 -0.4 ng/well of recombinant human PF- 4. Western Blot: To detect human PF - 4 by Western Blot analysis this antibody can be used at a concentra tion of 0.1 - 0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hPF-4 is 1.5 -3.0 ng/lane, under either reducing or non-reducing conditions. Immunohistochemistry: This antibody stained formalin-fixed, paraffin-embedded sections of human breast malignant ductal adenocarcinoma. The recommended concentration is 1.0 mg/mL -2.0 mg/mL with an overnight incubation at 4°C. An alkaline phosphatase-labeled polymer detection system was used with a red chromogen. Heat induced antigen retrieval with a pH 6.0 sodium citrate buffer is recommended. Optimal concentrations and conditions may vary.

PF-4 is a CXC chemokine that is expressed in megakaryocytes and stored in the alpha-granules of platelets. PF-4 is chemotactic towards neutrophils and monocytes and has been shown to inhibit angiogenesis. Recombinant human PF-4 is a 7.8 kDa protein containing 70 amino acid residues, including the four highly conserved residues present in CXC chemokines.
