Total cell lysates from HeLa (negative control, lane 1) and E. coli strain BL21 (lane 2) cells were resolved by electrophoresis (4-12% Bis-Tris), transferred to nitrocellulose, and probed with 0.1 ug/ml (1:5000 dilution) of purified RNA Polymerase subunit alpha (clone 4RA2). Proteins were visualized by chemiluminescence detection using HRP goat anti-mouse IgG antibody at a 1:3000 dilution. Ponceau S red staining (bottom) was used to confirm equal protein loading. Lane M is the molecular weight ladder.

Western Blot
other info1 :
Preparation: The antibody was purified by affinity chromatography.
other info2 :
Structure: alpha is a 36 kD subunit of the E. coli RNA polymerase.
products description :
DNA-dependent RNA polymerase catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates. This subunit plays an important role in subunit assembly since its dimerization is the first step in the sequential assembly of subunits to form the holoenzyme.

uniprot summary :
Function: DNA-dependent RNA polymerase catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates. This subunit plays an important role in subunit assembly since its dimerization is the first step in the sequential assembly of subunits to form the holoenzyme. HAMAP-Rule MF_00059. Catalytic activity: Nucleoside triphosphate + RNA(n) = diphosphate + RNA(n+1). HAMAP-Rule MF_00059. Subunit structure: Homodimer. The RNAP catalytic core consists of 2 alpha, 1 beta, 1 beta' and 1 omega subunit. When a sigma factor is associated with the core the holoenzyme is formed, which can initiate transcription. Domain: The N-terminal domain is essential for RNAP assembly and basal transcription, whereas the C-terminal domain is involved in interaction with transcriptional regulators and with upstream promoter elements. HAMAP-Rule MF_00059. Sequence similarities: Belongs to the RNA polymerase alpha chain family.
