Performance: Five x 10^5 cultured Raji human tumor cells were incubated 45 minutes on ice with 80 ul of anti-CD137L antibody at 10 ug/ml. Cells were washed twice and incubated with 2 degree reagent Goat anti-Mouse IgG/FITC, MBS666783 after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 0.29 log 10 fluorescent units when compared to a Mouse IgG2a negative control MBS666112 at a similar concentration.

Information: Human CD137 Ligand is a type II transmembrane protein constitutively expressed by monocytes, B cells neuroblastoma cells (1), and some activated T cells. The interaction of CD137 (4-1BB) and CD137L can supply a costimulatory signal to T cells through CD137. This costimulus seems to be more effective for CD8+ effector T cells than for CD4+ helper T cells (3). The interaction can also effect a reverse signal, inducing monocyte activation (2), or T cell inactivation and programmed death (4) through CD137L. Anti-CD137L antibody binds to CD137L present on Raji and U-937 cells, and also binds to recombinant CD137L-muCD8 in EIA. This antibody does not block the interaction between CD137 and CD137L.
