Assay Conditions: Paraoxonase-1 catalyses the cleavage of phenyl acetate (4 mM) in 20mM Tris HCl buffer pH 8, containing 1mM CaCl2 resulting in phenol formation. The rate of formation of phenol is measured by monitoring the increase in absorbance at 270nm at 25 C. One unit of arylesterase activity is equal to 1uM of phenol formed per minute. The activity is expressed in kU/L, based on the extinction coefficient of phenol of 1310 M-1 cm at 270nm, pH 8, and 25 C.
