Insulin-like Growth Factor 2 receptor (IGF-II R), also known as cation-independent mannose-6 phosphate receptor (CI-MPR), is a 300kD, 2264aa type I transmembrane glycoprotein of the P-type lectin family. The extracellular region contains 15 repeats that mediate homodimer formation and multifunctional binding of IGF-II and many other proteins that do and do not contain mannose-6 phosphates. IGF-II R generally regulates endocytosis and lysosomal/endosomal trafficking. Human IGF-II R shares 85% aa identity with mouse and cow IGF-II R within the extracellular region used as an immunogen.

The protein encoded by this gene is a receptor for both, insulin-like growth factor 2 (IGF2) and mannose 6-phosphate (M6P). The IGF2 and M6P binding sites are located on different segments of the receptor. This receptor functions in the intracellular trafficking of lysosomal enzymes, the activation of transforming growth factor beta, and the degradation of IGF2. While the mouse Igf2r gene shows exclusive expression from the maternal allele, imprinting of the human IGF2R gene appears to be polymorphic, with only a minority of individuals showing expression from the maternal allele.
