products description :
Enzymes of the phospholipase C family catalyze the hydrolysis of phospholipids to yield diacylglycerols and water-soluble phosphorylated derivatives of the lipid head groups. The phospholipase C gamma-2 (PLCgII) is an enzyme that plays a crucial role in intracellular signal transduction pathways. This enzyme is important because of its role in the generation of second messengers following the hydrolysis of phosphatidylinositol 4,5-bisphosphate. PLCgII has been implicated in collagen-induced signal transduction in platelets and antigen-dependent signaling in B-lymphocytes. It has been suggested that tyrosine kinases activate PLCgII. Btk-deficient B cells exhibited a marked reduction in phosphorylation of the Src homology 2 (SH2)-SH3 linker region sites, Tyr(753) and Tyr(759). Phosphorylation of both sites was restored by expression of Tec, but not Syk, family kinases. In contrast, phosphorylation of the PLCgamma2 carboxyl-terminal sites, Tyr(1197) and Tyr(1217), was unaffected by the absence of functional Btk.

ncbi summary :
Enzymes of the phospholipase C family catalyze the hydrolysis of phospholipids to yield diacylglycerols and water-soluble phosphorylated derivatives of the lipid head groups. A number of these enzymes have specificity for phosphoinositides. Of the phosphoinositide-specific phospholipase C enzymes, C-beta is regulated by heterotrimeric G protein-coupled receptors, while the closely related C-gamma-1 (PLCG1; MIM 172420) and C-gamma-2 enzymes are controlled by receptor tyrosine kinases. The C-gamma-1 and C-gamma-2 enzymes are composed of phospholipase domains that flank regions of homology to noncatalytic domains of the SRC oncogene product, SH2 and SH3.[supplied by OMIM]
uniprot summary :
PLCG2: a calcium dependent phosphatidylinositol-specific phospholipase C. The activated enzyme produces the second messenger molecules diacylglycerol and inositol 1,4,5-trisphosphate. It is a crucial enzyme in transmembrane signaling. Phosphorylated and activated by tyrosine kinases in response to signaling through a variety of growth factor receptors and immune system receptors. Protein type: Carbohydrate Metabolism - inositol phosphate; Phospholipase; EC 3.1.4.11; Motility/polarity/chemotaxis. Chromosomal Location of Human Ortholog: 16q24.1. Cellular Component: plasma membrane; cytosol. Molecular Function: signal transducer activity; protein binding; phospholipase C activity; phosphoinositide phospholipase C activity. Biological Process: platelet activation; inositol phosphate metabolic process; Wnt receptor signaling pathway; phospholipid catabolic process; calcium-mediated signaling; response to lipopolysaccharide; T cell receptor signaling pathway; activation of store-operated calcium channel activity; B cell receptor signaling pathway; inositol trisphosphate biosynthetic process; regulation of gene expression; release of sequestered calcium ion into cytosol; B cell differentiation; phosphatidylinositol biosynthetic process; negative regulation of programmed cell death; innate immune response; follicular B cell differentiation; blood coagulation.
