products description :
PERK, an eIF2 alpha kinase, is a transmembrane protein resident in the endoplasmic reticulum (ER) membrane and couples ER stress signals to translation inhibition. ER stress increases the activity of PERK, which then phosphorylates eIF2 alpha, resulting in reduced translation. PERK-deficient mice have defects in pancreatic beta cells several weeks after birth, suggesting a role for PERK-mediated translational control in protecting secretory cells from ER stress. PERK activation during ER stress correlates with autophosphorylation of its cytoplasmic kinase domain. Phosphorylation of PERK at Thr980 can serve as a marker for its activation status.

uniprot summary :
Function: Phosphorylates the alpha subunit of eukaryotic translation-initiation factor 2 (EIF2), leading to its inactivation and thus to a rapid reduction of translational initiation and repression of global protein synthesis . By similarity. Catalytic activity: ATP + a protein = ADP + a phosphoprotein. Enzyme regulation: Perturbation in protein folding in the endoplasmic reticulum (ER) promotes reversible dissociation from HSPA5/BIP and oligomerization, resulting in transautophosphorylation and kinase activity induction . By similarity. Subunit structure: Forms dimers with HSPA5/BIP in resting cells. Oligomerizes in ER-stressed cells . By similarity. Subcellular location: Endoplasmic reticulum membrane; Single-pass type I membrane protein . By similarity. Induction: By ER stress. Domain: The lumenal domain senses perturbations in protein folding in the ER, probably through reversible interaction with HSPA5/BIP . By similarity. Post-translational modification: Autophosphorylated . By similarity.N-glycosylated . By similarity. Sequence similarities: Belongs to the protein kinase superfamily. Ser/Thr protein kinase family. GCN2 subfamily.Contains 1 protein kinase domain.
