specificity :
Clone 3B3 is an antibody highly specific for alpha-skeletal actin, and does not cross react with other actin isoforms.
form :
Each vial contains 100 ul 1 mg/ml purified monoclonal antibody in PBS containing 0.09% sodium azide.
storage stability :
The antibody is shipped at ambient temperature and may be stored at +4°C. For prolonged storage prepare appropriate aliquots and store at or below -20°C. Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance or the concentration of the product.
tested application :
ELISA, Immunoblotting, Immunohistochemistry (Frozen), Immunohistochemistry (Paraffin), Western Blotting.
app notes :
3B3 is useful for immunohistochemistry on frozen and paraffin-embedded tissues, immunoblotting and ELISA. Optimal antibody dilutions should be determined by titRation; recommended range is 1:100 - 1:1000 for immunohistochemistry with avidin-biotinylated Horseradish peroxidase complex (ABC) as detection reagent, and 1:1000 - 1:5000 for immunoblotting appliCations.

other info1 :
Source Note: 3B3 is a Mouse monoclonal IgG1 antibody derived by fusion of NS0 Mouse myeloma cells with spleen cells from a BALB/c Mouse immunized with a peptide comprising the N-terminal nonapeptide of alpha-skeletal actin with an acetylated N-terminus (Ac-DEDETTALVC-COOH) coupled to keyhole limpet hemocyanin through the cysteine residue.
other info2 :
Reactivity Note: The epitope recognized by alpha-SM1 is highly conserved. The antibody therefore cross-reacts with many species including protochordates, lower craniates and mammals.
products categories :
Cytoskeletal, Cardiovascular, Cell differentiation, Cytoskeleton, Stem cell research
products description :
Among the six actin isoforms described in mammals, two are found in virtually all cells (beta- and gamma-cytoplasmic), two are detected in smooth muscle cells (alpha- and gamma-smooth muscle) and two are present in striated muscles, one predominantly in skeletal (alpha-skeletal) and one in cardiac (alpha-cardiac) muscle cells. These actin isoforms differ slightly in their N-terminus, but the sequence of each of these actins is highly conserved in higher vertebRates. Skeletal alpha-actin may also be expressed in the mammalian heart. Although during the late stage of fetal development alpha-cardiac actin becomes the main isoform in the myocardium, alpha-skeletal actin might still make up for almost half of its total striated muscle actin content.

ncbi summary :
The product encoded by this gene belongs to the actin family of proteins, which are highly conserved proteins that play a role in cell motility, structure and integrity. Alpha, beta and gamma actin isoforms have been identified, with alpha actins being a major constituent of the contractile apparatus, while beta and gamma actins are involved in the regulation of cell motility. This actin is an alpha actin that is found in skeletal muscle. Mutations in this gene cause nemaline myopathy type 3, congenital myopathy with excess of thin myofilaments, congenital myopathy with cores, and congenital myopathy with fiber-type disproportion, diseases that lead to muscle fiber defects. [provided by RefSeq, Jul 2008]
uniprot summary :
ACTA1: Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells. Polymerization of globular actin (G-actin) leads to a structural filament (F-actin) in the form of a two-stranded helix. Each actin can bind to 4 others. Interacts with TTID. Interacts (via its C-terminus) with USP25; the interaction occurs for all USP25 isoforms but is strongest for isoform USP25m in muscle differentiating cells. Belongs to the actin family. Protein type: Cell development/differentiation; Contractile; Cytoskeletal; Motility/polarity/chemotaxis. Chromosomal Location of Human Ortholog: 1q42.13. Cellular Component: extracellular space; sarcomere; stress fiber; actin filament; cytosol; striated muscle thin filament; actin cytoskeleton. Molecular Function: protein binding; myosin binding; structural constituent of cytoskeleton; ADP binding; ATP binding. Biological Process: skeletal muscle fiber adaptation; muscle contraction; response to steroid hormone stimulus; response to mechanical stimulus; muscle thin filament assembly; response to lithium ion; response to extracellular stimulus; skeletal muscle fiber development; cell growth; muscle filament sliding. Disease: Nemaline Myopathy 3; Myopathy, Congenital, With Fiber-type Disproportion
