Recombinant Interleukin-18 was lyophilized from 0.2 um filtered PBS solution pH 7.4.

The lyophilized protein in stable for at least 2 years from date of receipt at -20 degree C. Upon reconstitution, this cytokine can be stored in working aliquots at 2-8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detecable loss of activity. Avoid repeated freeze/thaw cycles.

Domain: IL1. Host Note: Optimized DNA sequence encoding Human Interleukin-18 mature chain was expressed in Escherichia Coli. Biological Activity: The ED(50) was determined by the dose-dependent proliferation of human PBMC co-stimulated with IL-12 cells was found to be in the range of 5ng/ml. Reconstitution: A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.

Endotoxin: Endotoxin content was assayed using a LAL gel clot method. Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug). Molecular Weight Note: Native human Interleukin-18, generated by the proteolytic removal of the signal peptide and propeptide,the molecule has a calculated molecular mass of approximately 18 kDa. Recombinant Interleukin-18 is a disulfide-linked monomer protein consisting of 158 amino acid residue subunits, migrates as an approximately 18 kDa protein under non-reducing and reducing conditions in SDS-PAGE.

Cytokines

IL18 is synthesized as a biologically inactive precursor protein that has limited biological activity. The proform of IL18 is processed by one of the caspases, Caspase-1, to generate the 18 kDa bioactive molecule. IL-18 is produced during the acute immune response by macrophages and immature dendritic cells. Interleukin-18 is expressed by a variety of immune and non-immune cells, including monocytes and macrophages. IL18 is one of the pro- inflammatory cytokines. The activities of IL18 appear to be species specific. An important function of IL18 is the regulation of functionally distinct subsets of T-helper cells required for cell mediated immune responses (Nakanishi et al, 2001). IL18 functions as a growth and differentiation factor for Th1 cells. IL18 up-regulates FAS ligand mediated cytotoxic activitiy of murine natural killer cells. IL-18 is part of a complex regulatory circuit involved in causing cell death by apoptosis.
