form :
Recombinant Rantes/CCL5 was lyophilized from a 0.2 um filtered PBS solution.
storage stability :
The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C. Upon reconstitution, this cytokine can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity. Avoid repeated freeze/thaw cycles.
other info1 :
Domain: IL8. Host Note: Optimized DNA sequence encoding Human Rantes/CCL5 mature chain was expressed in Escherichia Coli.
other info2 :
Endotoxin: Endotoxin content was assayed using a LAL gel clot method. Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug). Reconstitution: A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers. Molecular Weight Note: Native human RANTES/CCL5 is generated by the proteolytic removal of the signal peptide and propeptide, the molecule has a calculated molecular mass of approximately 8 kDa. Recombinant RANTES/CCL5 is a disulfide-linked monomer protein consisting of 69 amino acid residue subunits and migrates as an approximately 8 kDa protein under non-reducing and reducing conditions in SDS-PAGE.

products description :
RANTES belongs to the family of chemotactic cytokines known as Chemokines. RANTES is expressed by an early response gene. The synthesis of RANTES is induced by TNF-alpha and IL1-alpha.RANTES is chemotactic for T-cells, human eosinophils and basophils and plays an active role in recruiting leukocytes into inflammatory sites. RANTES also activates eosinophils to release, for example, eosinophilic cationic protein. It changes the density of eosinophils and makes them hypodense, which is thought to represent a state of generalized cell activation and is associated most often with diseases such as asthma and allergic rhinitis. RANTES also is a potent activator of oxidative metabolism specific for eosinophils.

uniprot summary :
Function: Chemoattractant for blood monocytes, memory T-helper cells and eosinophils. Causes the release of histamine from basophils and activates eosinophils. Binds to CCR1, CCR3, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant RANTES protein induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). The processed form RANTES(3-68) acts as a natural chemotaxis inhibitor and is a more potent inhibitor of HIV-1-infection. The second processed form RANTES(4-68) exhibits reduced chemotactic and HIV-suppressive activity compared with RANTES(1-68) and RANTES(3-68) and is generated by an unidentified enzyme associated with monocytes and neutrophils. Ref.3 Ref.8 Ref.10 Ref.11 Ref.12. Subcellular location: Secreted. Tissue specificity: T-cell and macrophage specific. Induction: By mitogens. Post-translational modification: N-terminal processed form RANTES(3-68) is produced by proteolytic cleavage, probably by DPP4, after secretion from peripheral blood leukocytes and cultured sarcoma cells. The identity of the O-linked saccharides at Ser-27 and Ser-28 are not reported in Ref.8. They are assigned by similarity. Polymorphism: The variant Phe-24 is an antagonist of the chemokine receptors CCR1 and CCR3. Sequence similarities: Belongs to the intercrine beta (chemokine CC) family. Mass spectrometry: Molecular mass is 7515 1 Da from positions 27 - 91. Determined by SELDI.
