CD4 is a 55 kDa transmembrane glycoprotein belonging to the immunoglobulin superfamily. In humans CD4 is expressed of peripheral T-cells, thymocytes, on macrophages and granulocytes. CD4 acts as a cellular adhesion molecule binding MHC class 2 molecules. It stabilizes the interaction of MHC class 2 restricted T- cells and antigen-presenting cells expressing an antigen in combination with a MHC class 2 molecule. Interactions of CD4 with MHC class 2 molecules are crucial during thymic development and subsequently for the function of single- positive CD4 (+), CD8 (-) T- lymphocytes. CD4 functions as an effector molecule for intracellular signal transduction. In T- lymphocytes CD4 has been found to be associated with tyrosine kinase lck which phosphorylates a number of other cell surface proteins and is involved probably in transducing the intracellular signal when these proteins are bound to their receptors.

The protein encoded by this gene is the CD3-epsilon polypeptide, which together with CD3-gamma, -delta and -zeta, and the T-cell receptor alpha/beta and gamma/delta heterodimers, forms the T-cell receptor-CD3 complex. This complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways. The genes encoding the epsilon, gamma and delta polypeptides are located in the same cluster on chromosome 11. The epsilon polypeptide plays an essential role in T-cell development.
