IGF-1 and IGF-2 are members of a protein family of related proteins with hormonal activities including nerve growth factor.Cell types responding to the two IGF factors include adipocytes, chondrocytes, epithelial cells, fibroblasts, glial cells, hepatocytes, muscle cells, and osteoblasts. The IGF-2 receptor, CD222, is a single-chain protein of 250 kDa. It is a high affinity binding protein for IGF-2 and shows very low affinity for IGF-1 and none for insulin. The cytoplasmic domain of the receptor does not possess intrinsic tyrosine kinase activity.

It is an imprinted gene, expressed only from the paternal allele, and epigenetic changes at this locus are associated with Wilms tumour, Beckwith-Wiedemann syndrome, rhabdomyosarcoma, and Silver-Russell syndrome. A read-through INS-IGF2 gene exists, whose 5' region overlaps the INS gene and the 3' region overlaps this gene.
