Amino acid transport across the plasma membrane is mediated by both sodium-dependent and sodium independent transport mechanisms (1). The recent advances in the molecular biology techniques let to discovery of several amino acid transporters that are homologues to the amino acid transport system describe for tissue culture cells and in membrane vesicle preparations (2). It has been shown that sodiumindependent amino acid transporter covalently interact with the heavy chain of a cell surface antigen 4F2 also known as CD98. Structurally, 4F2 compose of a 4F2 heavy chain (4F2hc), an integral membrane protein and a light chain that has intrinsic amino acid transport properties. Various amino acid transporters including transport type L, y+L and xc - has been shown to be the 2F4 light chains and assemble with 2F4hc for their functional amino acid transport activities (3-5). Recently a protein structurally related to amino acid transporter, with a characteristic 12 transmembrane domains (TMDs) has been cloned mouse brain that associates with 2F4hc for its small neutral amino acid transport activity (1). This transporter belongs to the LAT2 family, a component for system L transporter. The ASC-1 showed 65% identity to LAT2, 45% to LAT1, y+LAT1 and KIAA0245/y+LAT2 type transporters (6). The transporter ASC-1 is a member of the solute carrier family 7 and serves as a cationic amino acid transporter (y+ system). The ASC-1 protein has 530 amino acids (MW 63-64 kDa) is expressed in brain, lungs and intestine. It has two consensus phosphorylation sites (PKA and PKC) that are conserved in all y+ transport system. ASC1 transporter also transports with high affinity D and L-amino acid-related compounds and may involve in glutamenergic transmission in brain by modulating D-serine at glutamiergic synapse. ASC-1 forms disulfide dimmers with ASC transporter and SLC3A2/4F2hc proteins. The ASC-1-selective antibodies were generated against unique N and C-terminal peptides that are unique to ASC-1 protein. MyBioSource has generated specific rabbit anti-sodium independent amino acid transporter (ASC-1) polyclonal antibodies utilizing linear and cyclic peptide sequences. These antibodies have been fully characterized for cross reactivity with other members of the amino acid tranporter system. MyBioSource has produced antibodies to multiple epitopes on the same protein that will facilitate studies utilizing interspecies cross reactivity. MyBioSource provides ASC-1 Western blot positive control in "ready-to-use" SDS-PAGE sample buffer. The ASC-1 positive control appears as a diffuse band of 63-65 kDa and co-migrates with rat brain and lung sodium-independent amino acid transporter protein.
