Leukocyte Ig-like receptors (LILRs) are the mammalian ortholog of Paired Ig-like Receptors (PirB) receptors which are encoded by several genes and are expressed on various immune cells from myeloid and lymphoid lineages (1). LILRs are divided into two types of receptors: activating-type receptors and inhibitory-type. LILRB receptor genes are located on chromosomal region 19q13.4 which encode 19 genes encoding leukocyte-expressed receptors of the immunoglobulin (Ig) superfamily. LIRs contain either two or four Ig domains and fall into three classes: (i) those with cytoplasmic domains containing two, three, or four immunoreceptor tyrosine-based inhibitory motif-like motifs; (ii) those with a short cytoplasmic domain and no immunoreceptor tyrosine-based inhibitory motif-like motifs; and (iii) those with no transmembrane domain represented by a single LILR molecule that is presumably secreted. LILRB3 receptors are expressed in at least 3 variant isoforms (isoform 1, 2 and 3) with molecular weight around 76-78kDa. The amino acid sequences in the extracellular regions of eight of these receptors show between 63 and 84% identity to the prototypic LILR1. The LILRB3 selective antibodies were made against an epitope that corresponds to amino acids ranging from 615-632 from human LILRB3 protein. The antibodies are affinity purified on an immobilized antigen based affinity matrix. The isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage.
