Principle of the Assay The assay is based on the sandwich ELISA technique. A monoclonal antibody specific for a conformational epitope on assembled AAV 5 capsids is coated onto microtiter strips and is used to capture AAV 5 particles from the specimen. Captured AAV 5 particles are detected in two steps. First a biotin conjugated monoclonal antibody to AAV 5 is bound to the immune complex. In the second step streptavidin peroxidase conjugate reacts with the biotin molecules. Addition of substrate solution results in a color reaction which is proportional to the amount of specifically bound viral particles. The absorbance is measured photometrically at 450 nm. The kit control provided contains an AAV 5 particle preparation of intact capsids. It shows a typical standard curve when used in serial dilution. It allows quantitative determination of samples of an unknown particle titre (immunological titre) and the calibration of an inhouse AAV 5 preparation (e.g. infectious titre, transducing units, DNA dot blot titre). Background/Introduction: Adeno-associated virus 5 (AAV 5) is a ssDNA virus that is a topic of intense study in gene therapy especially for the treatment of cystic fibrosis where it shows long term expression. AAV 5 was isolated from a human genital site and has been used in several studies showing superior infection of lung, central nervous system, muscle, eye and arthritic joints when compared to other AAV serotypes. AAV 5 structure has been analyzed by X-ray crystallographic studies. Methods for the characterisation of AAV preparations currently include titration ELISA, real-time PCR, DNA dot blot, determination of transducing units, infectious center assay, SDS-PAGE and electron microscopy.
