SDS-gel electrophoresis (after reduction): A a, B b, g bands. Essentially free of Plasminogen and Fibronectin; Traces of von Willebrand factor may be present. Factor XIII is removed employing a modification of the method of McDonagh et al, Bichem Biophys Acta 446: 345ff (1976). Cross-linking of a and b chains: Complete (in the presence of Factor XIIIa, 20mM Calcium Chloride, and 5mM DTT).

Biological Significance: Fibrinogen (factor I) is a soluble plasma glycoprotein, synthesised by the liver, that is converted by thrombin into fibrin during blood coagulation. This is achieved through processes in the coagulation cascade that activate the zymogen prothrombin to the serine protease thrombin, which is responsible for converting fibrinogen into fibrin.

Biohazard: This product has been tested at serum donor level by FDA approved methods and found NEGATIVE for HIV I/II, and HCV antibodies and Hepatitis B surface antigen. It is recommened to observered full safety precautions when handling this material, since no testing method can completely assure the absence of all infectious agents.

Together with fibrinogen alpha (FGA) and fibrinogen beta (FGB), polymerizes to form an insoluble fibrin matrix. Has a major function in hemostasis as one of the primary components of blood clots. In addition, functions during the early stages of wound repair to stabilize the lesion and guide cell migration during re-epithelialization. Was originally thought to be essential for platelet aggregation, based on in vitro studies using anticoagulated blood. However, subsequent studies have shown that it is not absolutely required for thrombus formation in vivo. Enhances expression of SELP in activated platelets via an ITGB3-dependent pathway. Maternal fibrinogen is essential for successful pregnancy. Fibrin deposition is also associated with infection, where it protects against IFNG-mediated hemorrhage. May also facilitate the antibacterial immune response via both innate and T-cell mediated pathways.
