anti-mouse monoclonal antibody reacts with CD40 which is expressed on B-lymphocytes and on a subset of both CD4+ and CD8+ T cells in the adult (but not neonatal) spleen1. The level of CD40 on B cells is enhanced following activation buy LPS1. The interactions of CD40 with its ligand CD40L are responsible for generating humoral responses, germinal center formation, and generating B cell memory2. Stimulating cells with immobilized 3/23 mAb increases expression of CD86 (B7.2) on B cells1,3. Though the 3/23 mAb itself is a weak mitogen, it produces a synergistic proliferative B cell response with IL-4 or with mitogenic antibodies against surface Ig 1,3. Costimulation with 3/23 mAb and IL-4 rescues B cells from apoptosis induced by hypercross-linking of sIgM or sIgD3. This antibody is suitable for use in flow cytometry. Other applications of this antibody include IHC of acetonefixed frozen sections2.

The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor has been found to be essential in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. AT-hook transcription factor AKNA is reported to coordinately regulate the expression of this receptor and its ligand, which may be important for homotypic cell interactions. Adaptor protein TNFR2 interacts with this receptor and serves as a mediator of the signal transduction. The interaction of this receptor and its ligand is found to be necessary for amyloid-beta-induced microglial activation, and thus is thought to be an early event in Alzheimer disease pathogenesis.
