specificity :
Specific for the ~95k dynamin protein phosphorylated at Ser778. Labels the purified protein phosphorylated in vitro by cdk5 but not by PKC. Does not cross react with other purified substrates of cdk5 (e.g. amphiphysin and synapsin). Immunolabeling is blocked by ?-phosphatase treatment.
purity :
Affinity Purified (Prepared from sheep serum by affinity purification via sequential chromatography on phospho- and dephosphopeptide affinity columns.)
form :
100 ul in 10 mM HEPES (pH 7.5), 150 mM NaCl, 100 ug BSA per ml and 50% glycerol. Adequate amount of material to conduct 10-mini Western Blots.
storage stability :
For long term storage -20 degree C is recommended. Stable at -20 degree C for at least 1 year.
tested application :
Western Blot (WB)
app notes :
Quality Control: Western blots performed on each lot. WB: 1:1000
other info1 :
Antigen: Phosphopeptide corresponding to amino acid residues surrounding the phospho-Ser778 of rat dynamin. Immunogen Information: Synthetic phospho-peptide corresponding to amino acid residues surrounding Ser778 conjugated to KLH. Immunogen Species: Rat
other info2 :
Reactivity Assumed Based on 100% Sequence Homology: Bovine, canine, chicken, human and mouse. Species Reactivity Note: The antibody has been directly tested for reactivity in Western blots with rat tissue. It is anticipated that the antibody will react with bovine, canine, chicken, human and mouse based on the fact that these species have 100% homology with the amino acid sequence used as antigen. Biological Significance: Dynamin is a member of a group of nerve terminal proteins called dephosphins that regulate synaptic vesicle endocytosis (Cousin et al., 2001; Graham et al., 2002; Tsuboi et al., 2002). Cyclin dependent protein kinase 5 phosphorylates dynamin at Ser774 and Ser778 that are the phosphorylation sites on dynamin phosphorylated in vivo (Tan et al., 2003). Phosphorylation of these sites on dynamin is thought to play a key role in synaptic vesicle trafficking.

uniprot summary :
Function: Microtubule-associated force-producing protein involved in producing microtubule bundles and able to bind and hydrolyze GTP. Most probably involved in vesicular trafficking processes. Involved in receptor-mediated endocytosis . By similarity. Catalytic activity: GTP + H2O = GDP + phosphate. Subunit structure: Interacts with CAV1 and SH3GLB1. Interacts with SNX9. Interacts with SNX33 (via SH3 domain) . By similarity. Interacts with PHOCN. Interacts with PACSIN1, PACSIN2 and PACSIN3 . By similarity. Binds SH3GL1, SH3GL2 and SH3GL3. Interacts with MYO1E (via SH3 domain). Ref.6 Ref.7 Ref.10. Subcellular location: Cytoplasm Ref.8. Cytoplasm cytoskeleton. Note: Microtubule-associated. Isoform-specific localization. Ref.8Isoform 2: Cytoplasm Ref.8. Isoform 6: Cytoplasm Ref.8. Golgi apparatus Ref.8. Tissue specificity: Brain-specific (peripheral sensory neurons). Ref.8. Developmental stage: Expressed in neurons after maturation. Post-translational modification: Constitutively phosphorylated by CDK5 in nerve terminals and dephosphorylated by calcineurin when synaptic vesicle endocytosis (SVE) is stimulated by depolarization-dependent calcium influx.
