Intended Use The human IL-10 ELISA is an enzyme-linked immunosorbent assay for the detection of human IL-10. For research use only, not for use in diagnostic procedures. PRINCIPLE OF THE TEST An anti-human IL-10 coating antibody is adsorbed onto microwells. Human IL-10 present in the unknown or calibrator binds to antibodies adsorbed to the microwells. A biotin-conjugated anti-human IL-10 antibody is added and binds to human IL-10 captured by the first antibody. Following incubation unbound biotin-conjugated anti-human IL-10 antibody is removed during a wash step. Streptavidin-HRP is added and binds to the biotin-conjugated anti-human IL-10 antibody. Following incubation unbound Streptavidin-HRP is removed during a wash step, and substrate solution reactive with HRP is added to the wells. A coloured product is formed in proportion to the amount of human IL-10 present in the unknown or calibrator. The reaction is terminated by addition of acid and absorbance is measured at 450 nm. A calibrator curve is prepared from 7 human IL-10 calibrator dilutions and human IL-10 unknown concentration determined.

The protein encoded by this gene is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract. Mutations in this gene are associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis.[provided by RefSeq, May 2011]
