products description :
Principle of the Assay: An anti-human PSGL-1 coating antibody is adsorbed onto microwells. Human PSGL-1 present in the unknown or calibrator binds to antibodies adsorbed to the microwells. A biotin-conjugated anti-human PSGL-1 antibody is added and binds to human PSGL-1 captured by the first antibody. Following incubation unbound biotin-conjugated anti-human PSGL-1 antibody is removed during a wash step. Streptavidin-HRP is added and binds to the biotin-conjugated anti-human PSGL-1 antibody. Following incubation unbound Streptavidin-HRP is removed during a wash step, and substrate solution reactive with HRP is added to the wells. A colored product is formed in proportion to the amount of human PSGL-1 present in the unknown or calibrator. The reaction is terminated by addition of acid and absorbance is measured at 450 nm. A calibration curve is prepared from 6 human PSGL-1 calibrator dilutions and human PSGL-1 unknown concentration determined.

ncbi summary :
This gene encodes a glycoprotein that functions as a high affinity counter-receptor for the cell adhesion molecules P-, E- and L- selectin expressed on myeloid cells and stimulated T lymphocytes. As such, this protein plays a critical role in leukocyte trafficking during inflammation by tethering of leukocytes to activated platelets or endothelia expressing selectins. This protein requires two post-translational modifications, tyrosine sulfation and the addition of the sialyl Lewis x tetrasaccharide (sLex) to its O-linked glycans, for its high-affinity binding activity. Aberrant expression of this gene and polymorphisms in this gene are associated with defects in the innate and adaptive immune response. Alternate splicing results in multiple transcript variants.[provided by RefSeq, Apr 2011]
uniprot summary :
SELPLG: A SLe(x)-type glycan, which through high affinity, calcium-dependent interactions with E-, P- and L-selectins, mediates rapid rolling of leukocytes over vascular surfaces during the initial steps in inflammation. PSGL1 is critical for the initial leukocyte capture. Protein type: Membrane protein, integral. Chromosomal Location of Human Ortholog: 12q24. Cellular Component: membrane; integral to plasma membrane; integral to membrane; plasma membrane. Molecular Function: protein binding; receptor binding. Biological Process: blood coagulation; cell adhesion; leukocyte tethering or rolling; leukocyte migration; leukocyte adhesive activation
