Principle of the Assay: An anti-human PSGL-1 coating antibody is adsorbed onto microwells. Human PSGL-1 present in the unknown or calibrator binds to antibodies adsorbed to the microwells. A biotin-conjugated anti-human PSGL-1 antibody is added and binds to human PSGL-1 captured by the first antibody. Following incubation unbound biotin-conjugated anti-human PSGL-1 antibody is removed during a wash step. Streptavidin-HRP is added and binds to the biotin-conjugated anti-human PSGL-1 antibody. Following incubation unbound Streptavidin-HRP is removed during a wash step, and substrate solution reactive with HRP is added to the wells. A colored product is formed in proportion to the amount of human PSGL-1 present in the unknown or calibrator. The reaction is terminated by addition of acid and absorbance is measured at 450 nm. A calibration curve is prepared from 6 human PSGL-1 calibrator dilutions and human PSGL-1 unknown concentration determined.

This gene encodes a glycoprotein that functions as a high affinity counter-receptor for the cell adhesion molecules P-, E- and L- selectin expressed on myeloid cells and stimulated T lymphocytes. As such, this protein plays a critical role in leukocyte trafficking during inflammation by tethering of leukocytes to activated platelets or endothelia expressing selectins. This protein requires two post-translational modifications, tyrosine sulfation and the addition of the sialyl Lewis x tetrasaccharide (sLex) to its O-linked glycans, for its high-affinity binding activity. Aberrant expression of this gene and polymorphisms in this gene are associated with defects in the innate and adaptive immune response.
