Immunohistochemistry: In paraffin embedded Human Colon shows textured staining in the cytoplasm of absorptive cells. Recommended concentration, 4-6ug/ml. Western Blot: Preliminary experiments gave bands at approx 28kDa and 25kDa in lysates of Human Spleen and Tonsil and in Mouse Brain lysate after 1ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the bands we observe given the calculated size of 45.3kDa according to NP_002155.1. Both detected bands were successfully blocked by incubation with the immunizing peptide (and BLAST results with the immunizing peptide sequence did not identify any other proteins to explain the additional bands). Peptide ELISA: antibody detection limit dilution 1:8000.

This gene encodes indoleamine 2,3-dioxygenase (IDO) - a heme enzyme that catalyzes the first and rate-limiting step in tryptophan catabolism to N-formyl-kynurenine. This enzyme acts on multiple tryptophan substrates including D-tryptophan, L-tryptophan, 5-hydroxy-tryptophan, tryptamine, and serotonin. This enzyme is thought to play a role in a variety of pathophysiological processes such as antimicrobial and antitumor defense, neuropathology, immunoregulation, and antioxidant activity. Through its expression in dendritic cells, monocytes, and macrophages this enzyme modulates T-cell behavior by its peri-cellular catabolization of the essential amino acid tryptophan.[provided by RefSeq, Feb 2011]
