Immunohistochemistry: In paraffin embedded Human Testis shows granular cytoplasm staining in Leydig cells. Recommended concentration, 3ug/ml. Western Blot: Approx 120kDa band detected in HFF cell lysates. Data kindly provided by Laura Papp and Kum Kum Khanna. The observed molecular weight corresponds to earlier findings in literature with different antibodies (Papp et al, Mol Cell Biol. 2006 Jul;26(13):4895-910; PMID: 16782878). Recommended concentration: 0.1-0.3ug/ml. Peptide ELISA: antibody detection limit dilution 1:64000. The incorporation of selenocysteine into a protein requires the concerted action of an mRNA element called a sec insertion sequence (SECIS), a selenocysteine-specific translation elongation factor and a SECIS binding protein. With these elements in place, a UGA codon can be decoded as selenocysteine. The gene described in this record encodes a nuclear protein that functions as a SECIS binding protein. Mutations in this gene have been associated with a reduction in activity of a specific thyroxine deiodinase, a selenocysteine-containing enzyme, and abnormal thyroid hormone metabolism.
