Peptide ELISA: Antibody detection limit dilution 1: 64000. Immunohistochemistry: In paraffin embedded Human Testis shows textured cytoplasm staining in all germ cells. Recommended concentration, 4-6ug/ml. Western Blot: Preliminary experiments showed a 100kDa band but also bands at approx 35kDa and 26-28kDa in Human Ovary, Placenta, Prostate and Testis lysates after 0.1ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the additional bands we observe given the calculated size of 98.6kDa according to NP_004755.2. All detected bands were successfully blocked by incubation with the immunizing peptide (and BLAST results with the immunizing peptide sequence did not identify any other proteins to explain the additional bands).

PIWIL1: Plays a central role during spermatogenesis by repressing transposable elements and prevent their mobilization, which is essential for the germline integrity. Acts via the piRNA metabolic process, which mediates the repression of transposable elements during meiosis by forming complexes composed of piRNAs and Piwi proteins and govern the methylation and subsequent repression of transposons. Directly binds methylated piRNAs, a class of 24 to 30 nucleotide RNAs that are generated by a Dicer- independent mechanism and are primarily derived from transposons and other repeated sequence elements. Besides their function in transposable elements repression, piRNAs are probably involved in other processes during meiosis such as translation regulation. Probable component of some RISC complex, which mediates RNA cleavage and translational silencing. Also plays a role in the formation of chromatoid bodies and is required for some miRNAs stability.
