The lyophilized protein is stable for a few weeks at room temperature, but best stored at -20 degree C. Reconstituted human BDNF should be stored in working aliquots at -20 degree C.

Solubility: It is recommended to reconstitute the lyophilized hBDNF in sterile water or 0.4% NaHCO3 adjusted tp pH 8-9, not less than 100ug/ml, which can then be further diluted to other aqueous solutions, preferably in presence of carrier protein. Protein Content: Protein quantitation was carried out by UV spectroscopy at 280 nm using the absorbancy value of 0.95 as the extinction coefficient for a 0.1% (1mg/ml) solution at pH 8.0. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). Dimers and Aggregates: Over 95% dimer under non denaturing conditions

Endotoxin: Endotoxin level is less than 0.1 ng per ug (1EU/ug). Formulation: The protein was lyophilized from a concentrated (1mg/ml) solution with 0.02-0.03% NaHCO3. Biological Activity: Determined by its ability to stimulate chick E8 neurite outgrowth. The expected ED50 for this effect is 0.1-1.0 ng/ml.

BDNF is a member of the NGF family of neurotrophic growth factors. Like other members of this family, BDNF supports neuron proliferation and survival. BDNF can bind to a low affinity cell surface receptor called LNGFR, which also binds other neurotrophins such as NGF, NT-3 and NT-4. However, BDNF mediates its neurotrophic properties by signaling through a high affinity cell surface receptor called gp145/trkB. BDNF is expressed as the C-terminal portion of a 247 amino acid polypeptide precursor, which also contains a signal sequence of 18 amino acid residue and a propeptide of 110 amino acid residues. Recombinant human BDNF is a 27.0 kDa homodimer of two 119 amino acid subunits linked by strong non-covalent interactions.

The protein encoded by this gene is a member of the nerve growth factor family. It is induced by cortical neurons, and is necessary for survival of striatal neurons in the brain. Expression of this gene is reduced in both Alzheimer's and Huntington disease patients. This gene may play a role in the regulation of stress response and in the biology of mood disorders.
