The antibody NKI-C3 binds to a melanoma-associated antigen, which is mainly expressed in the cytoplasm of melanoma cells and naevus cellnaevi. The antibody doesn't react with normal melanocytes or other normal cells with the exception of mast cells, histiocytes, salivary gland, bronchial, pancreatic and prostate epithelia. To avoid True positive results in diagnosis the antibody should only be used in combination with non cross-reacting melanoma markers (e.g. HMB45 Art. No. ME100 and ME500). Melanocytes are pigmented Melanin containing cells, which are mainly located in the epidermis, protecting tissues from irradiation by sun beams (UVB). In melanoic tumours characteristic proteins are over-expressed, which in part are defined by specific antibodies. NKI/C3 in Western Blot stains diffuse protein bands in a range of 25 to 100 kDa from cytoplasm of melanoma cells 25-100 kDa peptide band in immuno blot of human melanoma.
