products description :
. Principle of the assay: This kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. The purified anti-IL-1betaantibody was pre-coated onto 96-well plates. And the HRP conjugated anti-IL-1beta antibody was used as detection antibodies. The standards, test samples and HRP conjugated detection antibody were added to the wells subsequently, mixed and incubated, then, unbound conjugates were washed away with wash buffer. TMB substrates (A & B) were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the IL-1beta amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of IL-1beta can be calculated. Background: Interleukin-1beta (IL-1beta) is a potent stimulator of bone resorption whose gene is mapped to 2q14, and has been implicated in the pathogenesis of high bone turnover and osteoporosis. It can reset glucose homeostasis at central levels. It cause oligodendrocyte death in coculture with astrocytes and microglia, and also can cause nuclear export of a specific NCOR corepressor complex, resulting in derepression of a specific subset of nuclear factor-kappa-B (NFKB)-regulated genes. Pathak et al. (2011) proposed that IL1B blockade may be a viable therapy for patients with autoimmune inner ear disease or sensorineural hearing loss that fail to respond to corticosteroids.

ncbi summary :
The protein encoded by this gene is a member of the interleukin 1 cytokine family. This cytokine is produced by activated macrophages as a proprotein, which is proteolytically processed to its active form by caspase 1. The encoded protein plays a role in thymocyte proliferation and is involved in the inflammatory response. [provided by RefSeq, Aug 2015]
uniprot summary :
Function: Produced by activated macrophages, IL-1 stimulates thymocyte proliferation by inducing IL-2 release, B-cell maturation and proliferation, and fibroblast growth factor activity. IL-1 proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells. Subunit structure: Monomer. Subcellular location: Secreted. Note: The lack of a specific hydrophobic segment in the precursor sequence suggests that IL-1 is released by damaged cells or is secreted by a mechanism differing from that used for other secretory proteins. Domain: The similarity among the IL-1 precursors suggests that the amino ends of these proteins serve some as yet undefined function. Sequence similarities: Belongs to the IL-1 family.
