Principle of the Assay: This kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. The purified anti-Progesterone receptor antibody was pre-coated onto 96-well plates. And the HRP conjugated anti-Progesterone receptor antibody was used as detection antibodies. The standards, test samples and HRP conjugated detection antibody were added to the wells subsequently, mixed and incubated, then, unbound conjugates were washed away with wash buffer. TMB substrates (A & B) were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Progesterone receptor amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Progesterone receptor can be calculated!!Background/Introduction: The progesterone receptor (PR, also known as NR3C3), is a member of the steroid receptor superfamily, mediates the physiologic effects of progesterone. It is activated by the steroid hormone progesterone. In humans, PR is encoded by a single PGR gene residing on chromosome 11q22, it has two main forms, A and B, that differ in their molecular weight. The human progesterone receptor exists as 2 functionally distinct isoforms, PRA and PRB. PRB functions as a transcriptional activator in most cell and promoter contexts, while PRA is transcriptionally inactive and functions as a strong ligand-dependent transdominant repressor of steroid hormone receptor transcriptional activity.
