Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-HGF polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-HGF polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the HGF amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of HGF can be calculated. Background/Introduction: Hepatocyte growth factor/scatter factor (HGF/SF) is a paracrine cellular growth, motility and morphogenic factor. It consists of 2 chains, heavy and light, with molecular mass of 54 to 65 kD and 31.5 and 34.5 kD, respectively. It is secreted by mesenchymal cells and targets and acts primarily upon epithelial cells and endothelial cells, but also acts on haemopoietic progenitor cells. It has been shown to have a major role in embryonic organ development, in adult organ regeneration and in wound healing. Human HGF plasmid DNA therapy of cardiomyocytes is being examined as a potential treatment for coronary artery disease as well as treatment for the damage that occurs to the heart after myocardial infarction. It may further play a role as an indicator for prognosis of chronicity for Chikungunya virus induced arthralgia.
