storage stability :
Store at 2-8 degree C. Do not freeze. The user must validate any other storage conditions. When properly stored, the reagent is stable to the date indicated on the label. Do not use the reagent beyond the expiration date. There are no definitive signs to indicate instability of this product; therefore, positive and negative controls should be tested simultaneously with unknown specimens. If unexpected results are observed which cannot be explained by variations in laboratory procedures and a problem with the reagent is suspected, contact Technical Support.
tested application :
Immunohistochemistry (IHC) Paraffin, Western Blotting (WB)
app notes :
Immunohistochemistry Procedure . Specimen Preparation: Formalin-fixed, paraffin-embedded tissues are suitable for use with this primary antibody. Deparaffinization: Deparaffinize slides using xylene or xylene alternative and graded alcohols. Antibody Dilution: If using the concentrate format of this product, dilute the antibody 1:100. The dilutions are estimates; actual results may differ because of variability in methods and protocols. Antigen Retrieval: Boil tissue section in 10mM citrate buffer, pH 6.0 for 10 min followed by cooling at room temperature for 20 min. Primary Antibody Incubation: Incubate for 30 minutes at room temperature. Slide Washing: Slides must be washed in between steps. Rinse slides with PBS/0.05% Tween. Visualization: Detect the antibody as instructed by the instructions provided with the visualization system. Immunohistochemistry Dilution: Ready To Use. IHC Positive Control: Breast Carcinoma, Mantle Cell Lymphoma. Western Blot . Recommended Starting Protocol: Dilute the antibody 1:25. Incubate for 1 hour at room temperature. The dilution is an estimate; actual results may differ because of variability in methods and protocols. Optimal dilution and procedure should be determined by the end user. Western Blot Positive Control: MCF7 Cell Lysate
other info2 :
Immunogen: Synthetic peptide from C-terminus of human cyclin D1. Control Tissue: Mantle Cell Lymphoma, Tonsil. Cellular Localization: Nucleus. Entrez Gene Code: 595. Pathway: Cell Cycle
products description :
Cyclin D1 or PRAD-1 or bcl-1 is one of the key cell cycle regulators, and functions in association with cdk4 and / or cdk6 by phosphorylating the Rb protein. It is a putative proto-oncogene overexpressed in a wide variety of human neoplasms including mantle cell lymphomas (MCL).

ncbi summary :
The protein encoded by this gene belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance throughout the cell cycle. Cyclins function as regulators of CDK kinases. Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event. This cyclin forms a complex with and functions as a regulatory subunit of CDK4 or CDK6, whose activity is required for cell cycle G1/S transition. This protein has been shown to interact with tumor suppressor protein Rb and the expression of this gene is regulated positively by Rb. Mutations, amplification and overexpression of this gene, which alters cell cycle progression, are observed frequently in a variety of tumors and may contribute to tumorigenesis.
