storage stability :
Store at 2-8 degree C. Do not freeze. The user must validate any other storage conditions. When properly stored, the reagent is stable to the date indicated on the label. Do not use the reagent beyond the expiration date. There are no definitive signs to indicate instability of this product; therefore, positive and negative controls should be tested simultaneously with unknown specimens. If unexpected results are observed which cannot be explained by variations in laboratory procedures and a problem with the reagent is suspected, contact Technical Support.

Immunohistochemistry (IHC) Paraffin, Western Blot (WB)
app notes :
Immunohistochemistry Procedure . Specimen Preparation: Formalin-fixed, paraffin-embedded tissues are suitable for use with this primary antibody. Deparaffinization: Deparaffinize slides using xylene or xylene alternative and graded alcohols. Antibody Dilution: If using the concentrate format of this product, dilute the antibody 1:100. The dilutions are estimates; actual results may differ because of variability in methods and protocols. Antigen Retrieval: Boil tissue section in 10mM EDTA for 10 min followed by cooling at room temperature for 20 min. Primary Antibody Incubation: Incubate for 30 minutes at room temperature. Slide Washing: Slides must be washed in between steps. Rinse slides with PBS/0.05% Tween. Visualization: Detect the antibody as instructed by the instructions provided with the visualization system. Immunohistochemistry Dilution: 1:100. IHC Positive Control: Hodgkin's Lymphoma. Western Blot . Recommended Starting Protocol: Dilute the antibody 1:100. Incubate for 1 hour at room temperature. The dilution is an estimate; actual results may differ because of variability in methods and protocols. Optimal dilution and procedure should be determined by the end user. Western Blot Positive Control: Ramos Cell Lysate

Immunogen: Synthetic peptide near C-terminus of human MUM1/IRF4. Control Tissue: Hodgkin's Lymphoma. Cellular Localization: Nucleus. Entrez Gene Code: 3662. Pathway: Transcriptional Regulators
products description :
The MUM1 (Multiple Myeloma Oncogene 1) gene was originally identified because of its involvement in the t(6;14) translocation observed in multiple myeloma, which causes juxtaposition of the MUM1 gene to the Ig heavy-chain locus. MUM1 is expressed in a wide spectrum of lymphoid neoplasms but not in myeloid and extra-hematopoitic tumors. MUM1 is a valuable marker for understanding and characterizing histogenesis of B-cell lymphomas. It is an excellent marker for Reed-Sternberg cells of classic Hodgkin's disease.

ncbi summary :
The protein encoded by this gene belongs to the IRF (interferon regulatory factor) family of transcription factors, characterized by an unique tryptophan pentad repeat DNA-binding domain. The IRFs are important in the regulation of interferons in response to infection by virus, and in the regulation of interferon-inducible genes. This family member is lymphocyte specific and negatively regulates Toll-like-receptor (TLR) signaling that is central to the activation of innate and adaptive immune systems. A chromosomal translocation involving this gene and the IgH locus, t(6;14)(p25;q32), may be a cause of multiple myeloma. Alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, Aug 2010]
uniprot summary :
IRF4: Transcriptional activator. Binds to the interferon- stimulated response element (ISRE) of the MHC class I promoter. Binds the immunoglobulin lambda light chain enhancer, together with PU.1. Probably plays a role in ISRE-targeted signal transduction mechanisms specific to lymphoid cells. Interacts with SPIB and DEF6. Not induced by interferons. Lymphoid cells. Belongs to the IRF family. 2 isoforms of the human protein are produced by alternative splicing. Protein type: DNA-binding; Oncoprotein; Transcription factor. Chromosomal Location of Human Ortholog: 6p25-p23. Cellular Component: nucleoplasm; membrane; cytoplasm; cytosol; nucleus. Molecular Function: protein binding; sequence-specific DNA binding; protein-lysine N-methyltransferase activity; transcription factor binding; transcription factor activity. Biological Process: T cell activation; transcription, DNA-dependent; cytokine and chemokine mediated signaling pathway; positive regulation of interleukin-2 biosynthetic process; positive regulation of transcription, DNA-dependent; regulation of T-helper cell differentiation; myeloid dendritic cell differentiation; defense response to protozoan; peptidyl-lysine methylation; positive regulation of interleukin-4 biosynthetic process; positive regulation of interleukin-10 biosynthetic process; positive regulation of interleukin-13 biosynthetic process; negative regulation of toll-like receptor signaling pathway; positive regulation of transcription from RNA polymerase II promoter; positive regulation of DNA binding. Disease: Skin/hair/eye Pigmentation, Variation In, 8
